Abstract

Aim of the study. To investigate the conditions for the biosynthesis of pectinesterase by the Trametes hirsuta strain IBK 1569 under submerged culture conditions.

Methodology. The effects of cultivation duration, lignocellulosic waste as a carbon source, its concentration and particle size, nitrogen source and concentration, initial medium pH, stirring rate and inoculum quantity on pectinesterase activity in the culture broth of T. hirsuta IBK 1569 were investigated. Enzyme activity was determined titrimetrically.

Scientific novelty. The peculiarities of the influence of the main parameters of deep cultivation on the synthesis of pectinesterase Trametes hirsuta IBK 1569 were established. The highest numerical value of PEA was recorded on the 9th day of cultivation, after which a tendency to its decrease was observed. Among the studied lignocellulosic wastes, the most effective source of carbon was beet pulp, while tangerine peels provided a statistically similar level of activity. The most favorable concentration of beet pulp was 10 g/dm3. The maximum numerical value of PEA was obtained for beet pulp particles with a size of 3–5 mm, however, no statistically significant differences were established in the range of 1–7 mm. High activity values were provided by urea, ammonium salts, potassium nitrate and peptone; for ammonium sulfate, the most favorable concentration was 1.5 g/dm3. The most favorable initial pH value was 6.0. A numerical decrease in PEA was observed already at 140 rpm, while at speeds of 160 rpm and above the activity was statistically significantly lower. The amount of seed material in the range of 2.5–10 % did not significantly affect PEA.

Conclusions. The results obtained demonstrate the feasibility of using beet pulp for the biosynthesis of pectinesterase by T. hirsuta IBK 1569 and may be utilised to optimise biotechnological processes for the production of pectinolytic enzymes.
